CONSTIPATION : A chronic condition that negatively impacts quality of life

Constipation is defined in human by episodes of fewer than three stools per week. Symptoms of constipation also include lower abdominal discomfort, a feeling of incomplete evacuation after a bowel movement, hard-to-pass stools, rectal bleeding, or anal fissure. Constipation is often a result of slow movement of material through the large bowel, resulting in a largely increased transit time. Constipation frequently alternates with diarrhea. Chronic constipation is a remarkably common and costly condition that can negatively impact the quality of life.

Models

We have developed and extensively characterized a subacute constipation model in rats induced by repetitive administration of loperamide (an anti-diarrhea compound) for chronic efficacy and MoA studies of compounds addressing Constipation. The model rapidly develops a decrease in total transit time associated with a strong decrease in fecal pellet output.
